By automated sequencing, no evidence for\n\t\t\t the presence of mutant PTEN transcripts was found. However, in\n\t\t\t situ hybridization on RA synovium revealed a distinct expression pattern\n\t\t\t of PTEN, with negligible staining in the lining layer but abundant expression\n\t\t\t in the sublining. Normal synovial tissue exhibited homogeneous staining for\n\t\t\t PTEN. In cultured RA-SF, only 40% expressed PTEN. Co-implantation of RA-SF and\n\t\t\t normal human cartilage into severe combined immunodeficiency (SCID) mice showed\n\t\t\t only limited expression of PTEN, with no staining in those cells\n\t\t\t aggressively invading the cartilage.
