Replicate light traps were deployed overnight from small boats in August of 2012, 2013, and 2014. Deployments were, ~5–6 m off the seafloor at all collection sites during the third quarter to new moon lunar periods. Simultaneous sampling across all sites was not possible. Larval fishes were removed from the samples and the remaining zooplankton were split using a Folsom Plankton Splitter or a box splitter. Raw counts of zooplankton were multiplied by the split denominator to determine zooplankton/trap. A total of two replicates for 19 nights of light trap deployments were identified for 29 major taxonomic zooplankton groups. These light trap deployments were conducted to sample nocturnal near-reef zooplankton to support project efforts to evaluate the coral ecosystem connectivity between shallow shelf coral reefs and mesophotic coral ecosystems. The project “Coral Ecosystem Connectivity: From Pulley Ridge to Florida Keys” is focused on investigating the role that the relatively healthy deep, light-dependent mesophotic coral ecosystems of Pulley Ridge may play in replenishing key fish species, such as grouper, and other organisms in the downstream reefs of the Florida Keys and Dry Tortugas. This interdisciplinary study is determining connectivity of specific reef species between Pulley Ridge and the Florida Keys, and describing the structure, and determining the value of Pulley Ridge’s mesophotic coral ecosystems. Because of the well-documented decline of Florida’s reefs, it is important to identify, protect, and manage sources of larvae that can help sustain Florida’s reef ecosystems and the tourism economy that depends on it. Light traps consisted of a ~1-m tall 505-μm mesh Nitex cylinder surrounding a white LED light. Traps were deployed overnight and three funnel-shaped openings in the cylinder enabled phototaxic nocturnal zooplankton to enter the traps and become trapped inside until trap retrieval the following morning (based on Sponaugle and Cowen 1996). Traps were deployed ~5-6 m off the seafloor across collection sites at Pulley Ridge (~67 m depth of seafloor), Dry Tortugas (~53 m depth of seafloor), and in the shallow (< 10 m depth of seafloor) and deep (20–30 m depth of seafloor) shelf in the lower Florida Keys (American Shoal and Looe Key). The collection sites were sampled in August 2012, 2013, and 2014 as follows: Pulley Ridge (2012 and 2014), Dry Tortugas (2013 and 2014), and the lower Florida Keys (2012 and 2013). Simultaneous sampling across all sites was not possible so samples processed focused on nights between the third quarter to new moon in the month of August. Note: light traps deployed at Pulley Ridge and Dry Tortugas were done in conjunction with a cruise on the R/V F.G. Walton Smith (Cowen 2015a, 2015b; Hanisak 2015). Upon retrieval, the contents of each trap were preserved in 95% ethanol and the sample brought back to the laboratory. All larval fishes were sorted from the samples and the remaining zooplankton were split using a Folsom Plankton Splitter or a box splitter. All zooplankton in the split subsample were identified to 29 major taxonomic groups. Some taxa (decapod crabs) were grouped by life stage: zoea vs megalopae.
