Samples were collected at six sites at various distances from a freshwater diversion in the vicinity of West Pointe a la Hache, Lake Hermitage, and Port Sulphur, Louisiana in May of 2018, 2019, and 2021. Five replicates were collected at each marsh location. TOC content, percent water, and grain size (percent Clay, Silt and Sand) are reported. Data are in spreadsheet format. These data will be included in a principal component analysis with other variables to investigate what parameters were most influential in the distribution of benthic macroinfauna in the study area, which will inform how river diversions influence ecological trajectory and functionality, as well as trophic relationships. Little is known about how river diversions influence the ecological trajectory, food web structure, and function of natural versus created marshes. To address this gap, this project established sites in the West Point a la Hache (WPH) area (Barataria Bay, in Plaquemines Parish, Louisiana), near the WPH siphon which periodically shunts Mississippi River water into the local marshes. Sites were also established in nearby marshes that were restored as part of the Lake Hermitage Marsh Creation Project, representing an ideal model system to examine how seasonal and spatial shifts in salinity due to river diversions influence species composition and food web structure in both natural and different-aged created marshes. Objectives of the overall project included characterizing species compositions and abundances in multiple trophic levels (microbes to upper trophic level predators) and applying bulk (SIA) and compound-specific stable isotope analysis (CSIA) of amino acids (AA) in producer and consumer tissues to describe and compare the structure and complexity of food webs and reveal aspects of fish residency. Sediment samples were collected at two locations at each site (10 and 50 meters from marsh edge) for both natural and restored marshes. Five replicates were collected at each location for a total of ten samples per site on a given sample date. Grain size Method: Sediment was collected with a 60 mL syringe corer at 10 meters and 50 meters from the marsh’s edge. Two plugs were taken with the corer and placed into a Whirl Pak bag, which was then placed into an ice chest. Once the samples were back at the laboratory and thawed, enough sample was added to fill up about 1/3 of a 50 mL centrifuge tube. The sediment was chemically treated with 5-10 mL of 30% Hydrogen Peroxide by covering the sample completely. The centrifuge tubes were then placed into a beaker filled with water on top of a hot plate at 80℃ to quicken the digestion process. Digestion of organic matter was continued until there was no more bubbling. Once the organic matter was completely digested, they were centrifuged at 4000 rpm for 4 minutes, which left a liquid surface layer. This top layer was removed via pipette and the remaining sample was covered with about 5mL of de-ionized water. The samples were then vortexed for 30 seconds and measured through a Beckmann-Coulter laser diffraction particle size analyzer (Model LS 13 320). TOC Method: Sediment was collected with a 60 mL syringe corer at 10 meters and 50 meters from the marsh’s edge. Two plugs were taken with the corer and placed into a Whirl Pak bag, which was then placed into an ice chest. Once the samples were back at the laboratory and thawed, about 10 grams of sediment were placed into a pre-weighed aluminum weigh dish. The weigh dish was pre-weighed with a plastic “lid” to prevent evaporation and the lid weight alone was also recorded with a Mettler AE 160 balance. The sample in the weigh dish was then placed on the balance with the lid situated on top and the “Wet Weight” was recorded in grams. Once all the samples were weighed, they were placed on a baking pan and into a drying oven at 60℃ for 72 to 96 hours. Once dried, the sediment was allowed to reach room temperature and then reweighed with the lid. Once all of the “dry weight” was recorded, the weigh dishes were placed into a muffle furnace. The set-point of the furnace was placed at 450℃ for 12 hours. After 12 hours the samples were allowed to cool to room temperature and reweighed with the lid to obtain the “combusted weight.” The weight of total organic carbon was derived from a percent based on loss on ignition.
