The study period includes two harmful algal bloom (HAB) events dominated by L. polyedra in 2011 and 2012, as well as a reference period without bloom conditions in 2013. Data were collected to evaluate the biogeochemical behavior of copper (Cu) and cadmium (Cd) during harmful algal blooms (HABs) of Lingulaulax polyedra in Todos Santos Bay. Seawater samples were filtered under a positive atmosphere in an ultra-clean laboratory using Nuclepore® polycarbonate filters (0.45 μm). Filters containing particulate material were stored at −8 °C until analysis. The filtered seawater was acidified to pH ~2 and dissolved Cu and Cd were pre-concentrated using Chelex-100 ion-exchange resin (Bio-Rad, Hercules, USA) following the methods of Bruland et al. (1979). Particulate Cu and Cd, retained on filters, were digested in Teflon vessels via microwave-assisted acid digestion system (ETHOS TC, Milestone, Freemont, USA), using HNO3 and H2O2 (OmniTrace), following the methods of Quevauviller et al. (1993). The digests were evaporated at 80 °C and re-dissolved in 10 mL of 0.2% HNO3 for analysis. Both fractions were quantified by graphite furnace atomic absorption spectrophotometry (GF-AAS; AAnalyst 600, Perkin–Elmer, Shelton, USA.; Data Quality Method: Accuracy and precision were verified using certified reference materials: CASS-5 (seawater, Canadian Research Council of Canada, Ottawa, Canada) and BCR 414 (plankton, Community Bureau of Reference, Geel, Belgium), with recoveries of 93–110% for Cud (n = 6) and 90–97% for Cup (n = 9) and a precision of 5–10%. Percent recovery for Cdd and Cdp were 93 ± 4% and 100 ± 10%, respectively.
